The discovery paper identified stomach ghrelin as the endogenous ligand for GHS-R1a (PMID 10604470; checked 24 Aug 2026). It found that octanoylation at serine 3 was required for activity.
How is Lenomorelin different from ghrelin mimetics?
This is the body's own acyl-ghrelin. Ipamorelin, GHRP-2, GHRP-6, and Hexarelin are synthetic molecules that act at the same receptor.
A result for a mimetic is not a Lenomorelin result. A Lenomorelin result does not apply to a mimetic.
What did the 16-person appetite study find?
The study was randomized, double-blind, and placebo-controlled (PMID 16247504; checked 24 Aug 2026). It had eight women and eight men.
They ate 5,076 ± 691 kJ with ghrelin and 4,230 ± 607 kJ with salt water (PMID 16247504). The 27% gap had p = 0.04.
Palatability scores were 81.1 ± 3.6 with ghrelin and 70.0 ± 4.4 with salt water (PMID 16247504). This gap had p = 0.03.
The Medical Research Council paid for grant G7811974. The abstract gives no side-effect result.
What did the eight-week cancer study find?
Thirty-one cancer patients who were losing weight joined a randomized double-blind study (PMID 20186829; checked 24 Aug 2026). Seventeen were in the high group. Fourteen were in the low group. All got ghrelin, so there was no placebo arm.
The high group had a clear rise in appetite scores. They lost less body fat, with p < 0.04 (PMID 20186829). Blood GH also fell, with p < 0.05.
Most other results did not differ. The abstract says no side effect was seen in the high group. It gives no event table.

What did the growth-hormone study also record?
Twelve healthy participants took part in a study with more than one level (PMID 11124868; checked 24 Aug 2026). Mean GH peaks at the two top levels were 69.8 ± 9.2 and 90.9 ± 16.9 µg/L.
Prolactin also rose with the level. Two people had heavy sweating at the top levels.

What does evidence tier B mean here?
Human studies measured appetite, food intake, GH, and metabolic outcomes. The studies were small or used specific patient groups. Tier B still fits (checked 24 Aug 2026).
NCT00681486 calls a closed ghrelin test phase 3 and lists an estimate of 30 people. It posts no results. PMID 20186829 calls the close 31-person paper phase 2. The page keeps the two phase labels apart.
Why is the EU status investigational?
CTIS trial 2024-517598-26-00 names LENOMORELIN as the test drug (CTIS record; checked 24 Aug 2026). The test product is Acyl Ghrelin 0.25 mg. Sweden approved the trial on 21 October 2024. The trial has now ended.
CTIS trial 2024-515705-26-00 also names LENOMORELIN as the test drug (CTIS record; checked 24 Aug 2026). The Netherlands approved it on 15 October 2024. It is still approved as a trial.
